Comet Assay

The Pathfinder™ system, complete with our Comet applied software module, allows fully automated comet and comet head detection, as well as automated quantification of the parameters of interest (total fluorescence, percentage of DNA in the tail, Head vs Tail fluorescence, Olive moment, Extent moment, etc). The Comet module automatically takes into account the halo around the head to provide accurate data.

Specific algorithms have been developed to accommodate a wide range of cell lines for in vitro comet assay. Concerning in vivo comet assay, dedicated algorithms for preparations from liver, bone marrow, blood, lung and stomach are available. Specific algorithms have also been designed to score comets from plant cells.

NEW: An integrated control box enables you to modify detection parameters and accommodate prospective research samples or sample prepared under new conditions.

The whole process is fully unattended, from image capture to export of final results. Since it is high-content imaging, all information on each individual comet is accessible, and this on thousands of comets. Integrated statistical tools (multi-histograms and scatter plots) enable fast and easy comparison of different sample on one graph. Selection of any sub-population on those graphs results in instant presentation of all comets belonging to those sub-populations, on the screen and in the eyepiece of the microscope.

Using ACCELL SFD robotic device, up to 200 samples can be fully automatically processed in one batch without human intervention. A single mouse click enables capture of the samples, detection analysis of thousands of comets and statistical analysis of different experimental conditions on a single plot.

Pathfinder™ technology enables a 32-fold increase in the number of slides processed per day as compared to conventional software-assisted scoring. Since Pathfinder scoring occurs without human intervention, it frees human work forces and it is 100% repeatable, reliable and operator-independent.

Recent developments for high-throughput parallelized comet assay experiments using Gelbond™ films are fully compatible with Pathfinder™ technology. IMSTAR is part of COMICS European program for empowering comet assay capabilities and throughput.
