Rare events detection represents a stumbling block for visual scoring. Automated scoring rapidly becomes necessary at a frequency of 1 to 1000, but the presence of artefacts renders its use difficult due to the detection of hundreds of false positives. IMSTAR Pathfinder’s high-resolution Rare Events detector enables rapid scoring of tens of thousands of cells. Thus, rare events can be quantified with suitable statistics. Depending on the quality of the preparation, the proportion of false positive signals can be as low as 0.1% as compared to the total number of detected objects. Automated relocation of rare events on the screen or in the eyepiece of the microscope enables visual inspection and instant validation of true-positive objects.
In the example above, male contaminant cells are present in a female cell line. X-chromosomes are labeled green, and Y-chromosomes in orange by co-localization of a probe labeled red and green. IMSTAR's Pathfinder™ with CellRarity Applied Software Module scored over 40.000 cells and presented only 65 rare event candidates for visual inspection. 9 of them were scored as true positives, and 4 were suspicious cases (images courtesy of Dr. Gallagher, Pfizer).
